Key responsibilities
- Peptide synthesis: Execute solid-phase peptide synthesis (SPPS) and liquid-phase peptide synthesis (LPPS) protocols reliably and efficiently. Plan and carry out sequences of deprotection, coupling, and washing steps with careful attention to yield and purity.
- Reaction monitoring: Monitor progress of peptide reactions using qualitative and colorimetric tests (Kaiser, Chloranil, Elman’s test) and interpret chromatographic results to guide reaction optimization.
- Preparative purification: Operate and optimize preparative HPLC separations using DAC columns and other preparative media to isolate target peptides at research and pilot scales.
- Analytical support: Perform routine analytical HPLC for purity assessment and support method development for preparative purification. Prepare samples for LC–MS and interpret mass spectra.
- Characterization: Characterize synthesized peptides using IR, mass spectrometry (LC–MS/HRMS), and basic NMR interpretation as needed to confirm sequence and structural features.
- Equipment operation & maintenance: Operate and maintain critical R&D equipment, including flash chromatography systems, rotary evaporators, nanofiltration units, and lyophilizers. Ensure equipment is calibrated and maintained per SOPs.
- Documentation & compliance: Maintain accurate experimental records and batch documentation according to GLP/GMP-style standards. Prepare technical reports and analytical summaries suitable for client review.
- Scale-up contribution: Support scale-up activities by transferring optimized lab-scale protocols to pilot or small-batch conditions, troubleshooting process inefficiencies, and assisting in technology transfer to manufacturing teams.
- Quality & safety: Follow laboratory safety practices, chemical handling procedures, and waste disposal protocols. Participate in quality checks, internal audits, and continuous improvement initiatives.
Essential qualifications & experience
- Education: M.Sc. in Organic Chemistry. Specialization or thesis work in peptide chemistry is strongly preferred.
- Experience: 4–6 years of demonstrable hands-on experience in peptide synthesis (SPPS and LPPS) and purification in a CRO, CDMO, pharmaceutical R&D or academic research environment.
- Technical skills:
- Proficient with SPPS workflows, peptide coupling strategies, and protecting group chemistry.
- Strong experience in preparative HPLC purification and analytical HPLC method support.
- Familiarity with nanofiltration and lyophilization for peptide concentration and isolation.
- Competent in interpreting LC–MS data, IR spectra, and basic NMR peaks relevant to peptide characterization.
- Experience in handling DAC columns and flash chromatography for peptide purification.
- Documentation & GLP: Proven ability to maintain GLP-style documentation and adhere to SOPs and quality standards in an R&D environment.
